A série enfoca a dinâmica entre os colegas de apartamento, os físicos Leonard e Sheldon, e a nova vizinha do prédio, a bela Penny, que desperta a atenção de Leonard à primeira vista. Leonard e Sheldon recebem frequentemente a visita de seus dois amigos e também colegas de trabalho na Universidade, Rajesh e Howard, e juntos usam o tempo livre para todo tipo imaginável de divertimento nerd: de palavras cruzadas em idioma Klingon, passando por maratonas de séries de ficção científica às sagradas noites de quarta jogando Halo. Penny aos poucos vai se integrando a esse estranho novo ambiente, onde ouve discussões sobre a física do super-homem e mecânica quântica, enquanto os intelectuais são apresentados a novos conceitos, como o amor.
